Question: How long will it take to generate Gene and Protein Expression libraries?
Answer: Below are approximately how long it will take to perform the sample preparation and workflow steps.
Tissue Preparation:
- Post-sectioning, tissue is dried for 3 hours at 42°C, and can then be stored in a room temperature desiccator overnight prior to starting the staining protocol.
- Deparaffinization includes baking the tissue at 60°C for 2 hours and performing tissue hydration for ~1 hour.
- Tissue staining and Decrosslinking will be variable depending on the staining protocol performed:
- H&E staining and Decrosslinking will take ~1.5 hours
- Decrosslinking and direct IF staining will take ~2.5 hours
- Decrosslinking and indirect IF staining will take ~3.5 hours
Workflow:
- The workflow occurs over three days (post-tissue baking). The first day includes staining and Hybridization. The second day includes antibody labeling. The third day includes completing CytAssist-mediated probe and antibody tag transfer, on-slide extension, and off-slide library construction steps.
- If done continuously, the first day is estimated to take ~5-7.5 hours (depending on the staining method performed), the second day is estimated to take ~2 hours, and the third day is expected to take ~7 hours.
