Answer: The following thermal cyclers have been validated by 10x Genomics for use with Xenium In Situ Gene Expression as outlined on the support webpage thermal cycler compatibility table.

Here is a list of thermal cyclers that were tested internally and found to be incompatible with Xenium v1. Please note, failure modes listed below are usually intermittent. For example, slides might only crack ~20% of the time. Reduced sensitivity may not impact every run or the entire tissue section.

For all unsupported models, we cannot guarantee assay performance or offer replacements for unexpected gene expression or cell segmentation staining for experiments run on these. Unsupported thermal cyclers may be incompatible with the Xenium Slide Cassettes, leading to cassette leakage or evaporation, broken slides, and reduced assay sensitivity as compared to validated thermal cyclers. These phenotypes are most often observed during overnight incubation and high-temperature steps.
