Question: What alternative TE buffers can be used for Xenium?
Answer: TE Buffer, TRIS-EDTA, 1X Solution, pH 8.0 (Fisher Scientific, BP24731) is an important component of the post-Amplification wash. The EDTA in TE buffer helps with the inactivation of enzymes. The listed item has been tested by 10x Genomics and performs optimally with the assay.
If the recommended part number cannot be obtained for the In Situ Gene Expression workflow, the following buffers are considered low-risk alternatives:
- Promega (V6231); 100ml; TE Buffer, 1X, Molecular Biology Grade
- Invitrogen (AM9849); 500 ml; TE, pH 8.0, RNase-free
- FujiFilm Wako (314-90021); 100 ml; TE (pH 8.0)
Additional TE buffers may also be low-risk as long as they are molecular biology grade, nuclease-free, equivalent EDTA molarity, and the same pH. We recommend purchasing TE buffer from a vendor and not preparing the buffer from reagents in the lab.
Substituting additional materials not listed above may adversely affect system performance. Lower levels of EDTA (e.g. TE with 0.1mM EDTA instead of 1mM EDTA) will negatively impact assay performance and are not recommended.
